Is it a legitimate request related to Duo to change phone passcode ?

I’m not quite sure what you’re asking. If a Duo administrator sets a policy requiring screen lock then Duo users who have Duo Mobile installed may need to configure screen lock before authenticating.

If the Duo Admin requires full-disk encryption then Android users need to configure a PIN for unlocking the device at boot.
